MACSelect™ Vectors and Tag Vector Sets
Streamlined protein expression and isolation in mammalian cells - MACS® Technology overcomes low transfection efficiencies and small amounts of protein.

The MACSelect™ - Transfected Cell Selection System overcomes low transfection efficiencies by magnetic enrichment of transfected mammalian cells. A cell surface marker, which can be specifically labeled with MACSelect MicroBeads for magnetic cell isolation, is coexpressed with the gene-of-interest. pMACS vectors encode the MACSelect surface markers CD4, H-2Kk, or LNGFR, and can be used for cotransfection or cloning (for further information on the MACSelect Systems, please refer to MACSelect - Transfected Cell Selection Kits).

Beside low transfection efficiencies, small amounts of expressed target proteins can be a challenge when working with mammalian cells. Especially for sensitive analyses, such as protein interaction studies, a highly specific and gentle purification is necessary. By expression of recombinant proteins with an epitope tag, target proteins can be isolated with high purity and sensitivity by using µMACS™ Tag Isolation Kits. These make use of paramagnetic MicroBeads coupled to highly specific, monoclonal antibodies binding His-, c-myc-, HA-, GST- or GFP-tagged proteins (please refer to µMACS Tag Protein Isolation Kits for detailed information). To streamline transfected cell enrichment and protein isolation, pMACS Kk vectors are now available with c-myc-, His-, or HA-tag sequences. With a single cloning step, the gene-of-interest is introduced into a pMACS Kk.Tag vector, which now encodes a surface marker for selection and the epitope tagged target protein. After transfection, positive cells are enriched and the recombinant proteins are isolated with fast and gentle MACS Technology.

Each MACSelect Vector Set includes two pMACS vectors for N- or C-terminal epitope tagging. pMACS Kk.Tag vectors are available with c-myc-, His-, or HA-tag sequences, for N- or C-terminal epitope-tagged protein expression from the CMV promoter. The mouse H-2Kk MACSelect Enrichment marker is coexpressed from the H-2Kk promoter (see fig. 1). The standard MACSelect Cotransfection and cloning vectors are available with the MACSelect Kits but can also be purchased separately.
